Scavenger resins
The role of palladium as a catalyst in organic chemistry has been well documented, including its use in such common reactions as Suzuki-style cross-coupling reactions, Heck reactions, and Buchwald amidations, as well as hydrogenations. Although palladium is recognized as an excellent catalyst, removing it from the target product is sometimes a problem, and for use in drug discovery, the active compound must contain palladium levels below a few parts per million. Traditional methods of removing palladium involve chromatography, but this can be labor- and solvent-intensive.

One modern technique for purifying reactions in drug discovery is the use of polymer-bound scavenger reagents. Two new palladium scavenger resins have been developed with which greater than 99% removal of palladium has been demonstrated by simply stirring either of them with a solution of the compound of interest at room temperature for 4–24 h.

A typical scavenging procedure is to add 2.2 molar equivalents of either PS-2-mercaptonicotinamide (1) or PS-dithiothreitol (2) to the target molecule dissolved in a tetrahydrofuran (THF) solution. The suspension is either agitated or stirred for 4–24 h and then filtered. The polymer reagent is washed twice with THF and filtered. The combined THF filtrates are then concentrated in vacuo to yield the final product. Test reactions used 22 mg of palladium acetate [Pd(OAc)2] and 220 mg of scavenging resin. After 4–24 h of agitation, less than 1 mg of mass was recovered. When tetrakis(triphenylphosphine)palladium(0) [Pd(Ph3P)4] was used as the palladium source, only triphenylphosphine [Ph3P] was recovered as determined by 1H-NMR spectroscopy and elemental analysis.

Micro parallel LC system
The Veloce micro parallel liquid chromatography (mPLC) system provides increased sample analysis capacity to scientists who need information to make timely research decisions. It uses established HPLC techniques, integrating them with conventional laboratory workflow to increase time and cost efficiency. The Veloce system is designed for fast, high-quality sample analysis in analytical chemistry, specifically in the drug discovery and development process.

The features of the Veloce system include increased sample analysis capacity and the ability to generate chromatograms comparable to those produced by conventional HPLC instrumentation for a broad class of analytical chemistry applications. The Veloce system provides up to 24 simultaneous separations. This parallel analysis provides the ability to analyze a large number of compounds simultaneously, generate a standard curve, and increase the number of replicates or conditions used in a study.

At the heart of the Veloce system is the 24-column Brio cartridge, which enables multiple samples to be analyzed in parallel for increased throughput. The cartridges can incorporate columns of varying lengths and a range of stationary phases. Researchers can select cartridges to fulfill throughput and to optimize performance for specific experimental protocols. Because the Veloce system is similar to traditional HPLC systems, scientists can continue to use their existing processes and protocols when adopting this product.

Reagents. The Qtracker 655 Kit is designed for loading Qdot nanocrystals into live cells. The kit uses custom targeting peptides to deliver highly luminescent Qdot quantum dots to the cytoplasm. Once inside the cells, Qtracker labels provide strong, stable fluorescence that does not bleach over time, even after extensive continuous illumination and observation. Qtracker quantum dots add no additional autofluorescence, are not transferred to adjacent cells in the population, and display excellent retention properties; the nanocrystals used in the kit remain inert and nontoxic to many cell types and can be followed in some cell lines for up to a week.
