Scavenger resins
The role of palladium as a catalyst in organic chemistry has been well documented, including its use in such common reactions as Suzuki-style cross-coupling reactions, Heck reactions, and Buchwald amidations, as well as hydrogenations. Although palladium is recognized as an excellent catalyst, removing it from the target product is sometimes a problem, and for use in drug discovery, the active compound must contain palladium levels below a few parts per million. Traditional methods of removing palladium involve chromatography, but this can be labor- and solvent-intensive.

One modern technique for purifying reactions in drug discovery is the use of polymer-bound scavenger reagents. Two new palladium scavenger resins have been developed with which greater than 99% removal of palladium has been demonstrated by simply stirring either of them with a solution of the compound of interest at room temperature for 4–24 h.

A typical scavenging procedure is to add 2.2 molar equivalents of either PS-2-mercaptonicotinamide (1) or PS-dithiothreitol (2) to the target molecule dissolved in a tetrahydrofuran (THF) solution. The suspension is either agitated or stirred for 4–24 h and then filtered. The polymer reagent is washed twice with THF and filtered. The combined THF filtrates are then concentrated in vacuo to yield the final product. Test reactions used 22 mg of palladium acetate [Pd(OAc)2] and 220 mg of scavenging resin. After 4–24 h of agitation, less than 1 mg of mass was recovered. When tetrakis(triphenylphosphine)palladium(0) [Pd(Ph3P)4] was used as the palladium source, only triphenylphosphine [Ph3P] was recovered as determined by 1H-NMR spectroscopy and elemental analysis.
